We have an Access Administrator that manually defines new users (and their specific roles) in Web Viewer.
Additionally, we have removed ALL repositories from the Default User role. However, some users (who have acquired their RACF IDs), are attempting to log into the Web Viewer prior to their 'role' assignment. This results in a Help Desk call to report that they cannot access any repository. To circumvent this, is it possible to disable the Default User 'assignment' that occurs automatically for 1st time users? Or, is it feasible (or advisable) to simply delete the Default User role entirely?

There is no way to disable the Default User role assignment for first-time users. And it is NOT advisable to delete the Default User role. The workaround is to have an administrator add the new user before they signon to Web Viewer for the first time.
