To utilize Bypass Ports for Network Activity functionality on Windows as per below documentation,

do we need to reboot Windows OS after the configuration? Or restarting PIM service will be enough to reflect the configuration?

Windows OS needs to be rebooted for the bypass network configuration to be reflected. This is because the changes will be picked up only during the load of the drivers.
