Issue/Introduction

The SAP Agent log files show the following error message: 

U02004172 Error while calling function module 'RFC_SYSTEM_INFO'
Initialization of repository destination _E failed: Connect from SAP gateway to RFC server failed

LOCATION    CPIC (TCP/IP) on local host with Unicode
ERROR       partner 'IP ADDRESS:PORT' not reached
TIME        DATE TIME
RELEASE     721
COMPONENT   NI (network interface)

Resolution

Most likely cause is an issue with the firewall/router/load balancer or other active network device. This can only be diagnosed by network traffic analysis and SAP Admin, and is out of Supports' scope.
